This prospective cohort study established patient-derived CRC organoids (75% success rate) and tumor-infiltrating lymphocytes (TILs, 87.5% success rate) as preclinical models for therapy prediction. Targeted NGS sequencing revealed TP53, APC, and KRAS as the most frequent mutations; MSI was detected in 11% of patients. Organoid-TIL co-culture enables exploration of the tumor microenvironment and immune evasion mechanisms, supporting personalized immunotherapy strategies. %L 500 %9 theses %9 Text %9 Hochschulschrift %R 10.18453/rosdok_id00005813 %U https://purl.uni-rostock.de/rosdok/id00005813 %U https://nbn-resolving.org/urn:nbn:de:gbv:28-rosdok_id00005813-8 %U https://d-nb.info/1420686658/34 %U https://doi.org/10.18453/rosdok_id00005813
